Mice were divided into three groups: normal control group, hepatic fibrosis group, and HT-treated group. An in vitro model was established by treatment with 5 ng/mL TGF-β1 for 72 h in L-02 hepatocyte cell line and in 2 × 10(6) fibroblasts cell line, and a mouse model of hepatic fibrosis was induced by carbon tetrachloride (CCl4) treatment. Administration of HT effectively attenuated liver injury by increasing serum levels of albumin and aminotransferase, and decreased liver tissue levels of α-smooth muscle actin (α-SMA) and collagen I. A reduction of transforming growth factor-beta1, phosphorylated signal transducer and activator of transcription 3 (p-STAT3) and mitogen-activated protein kinases (MAPKs) in the nucleus was observed in the HT-treated group. HT suppressed the production of collagen in vitro and in vivo and inhibited the TGF-β1-induced expression of fibronectin, α-SMA, and collagen I. Furthermore, HT suppressed the phosphorylation of Smad2/3 in the nucleus in a mouse model of hepatic fibrosis.
